CD40 在癫痫大鼠大脑皮层及海马中的表达变化

摘    要:目的:研究CD40炎症分子在癫痫持续状态(status epilepticus,SE)后大鼠大脑皮层及海马的表达。方法:采用免疫组织化学及免疫荧光双标记方法,观察锂-匹罗卡品癫痫大鼠大脑皮层及海马的不同区域、不同时间点、不同细胞中CD40的表达情况。结果:癫痫发作显著增加了CD40阳性细胞,以在海马组织中增加为著; SE后CD40主要在激活的小胶质细胞上表达;CD40阳性细胞在SE后3 d增加达到一个高峰,在SE 7 d后恢复到SE前稍高的水平。结论:CD40在激活的小胶质细胞上高表达,促进了SE大鼠海马炎症反应,提示CD40介导的信号通路参与SE后海马组织的炎症病理过程。

Abstract:Objective: To investigate the expression of inflammatory molecule CD40 in the pallium and hippocampus of rats aft er status epilepticus (SE).Methods: The expression of CD40 in the pallium, the different areas of hippocampus and the diff erent cells from the lithium-pilocarpine epileptic rats at diff erent time points were examined byimmunohistochemistry and double-immunofl uorescent labeling.Results: Aft er SE, CD40 expression was obviously inhibited, especially in hippocampus. CD40 wasmainly expressed in the activated microglia. CD40 positive cells reached a peak at the 3rd day andreturned to a slightly higher level at the 7th day aft er SE compared with the level before SE.Conclusion: Elevation of CD40 expression in the activated microglia can promote infl ammatory injury of rat’s hippocampus, suggesting that CD40 induced-signal pathway is involved ininflammatory injury in the hippocampus after SE.
