摘    要:目的 了解慢性阻塞性肺疾病(COPD)大鼠肺组织中的DCsmRNA表达变化.方法 通过烟熏+脂多糖(LPS)方法建立COPD大鼠模型.HE染色观察气道及肺组织病理学的改变,同时用荧光定量RT-PCR检测CD86mRNA及OX-62mRNA在肺组织中的表达变化.结果 COPD模型组大鼠肺组织HE染色符合慢性支气管炎及肺气肿的病理改变.与正常对照组相比,COPD模型组大鼠CD86mRNA及OX-62mRNA表达均增加(P<0.01).结论 COPD模型组大鼠第28天病理表现符合人类COPD的特征性病理改变,提示COPD大鼠模型建立成功.COPD模型组大鼠肺组织中CD86mRNA及OX-62mRNA的表达较正常对照组明显升高,提示树突状细胞(DCs)在COPD大鼠肺部表达增加,这可能是COPD发生发展的重要因素.

Abstract:Objective To study the expression of Dendritic Cells (DCs) mRNA in the lung tissue of rats with Chronic Obstructive Pulmonary Disease (COPD). Methods COPD was induced by established cigarette smoke inhalation and intratracheal LPS solution application. The pathomorphological changes in the lung were observed by hematoxylin - eosin staining (HE). The expressions of CD86 mRNA and OX - 62 mRNA in lung tissue were observed by RT - PCR. Results The changes of pathology of the COPD model group at 28 - day mimicked the pathology of COPD patients. The expression of CD86 mRNA and OX- 62 mRNA in the lung tissue were significantly increased in COPD model group, compared with normal control group ( P 〈 0.01 ). Conclusion The COPD rat model was established successfully by this combined methods. The expressions of DCs mRNA were increased in the lungs of rats with COPD. COPD is associated with creased numbers of DCs.
