大鼠肝贮脂细胞及Kupffer细胞的分离培养和鉴定

摘    要:
目的 肝贮脂细胞是肝纤维化发生机制研究的细胞学热点,该细胞的分离、培养具有重要意义。 方法 参考Friedman等的不连续密度梯度离心法,首次采用高分子聚蔗糖、泛影葡胺作为不连续密度梯度分层液分离细胞。 结果 本法成功地分离得到肝贮脂细胞和Kupffer细胞。 结论 本法经济、简便、可靠,细胞纯度高。

Abstract:
AIM Hepatic lipocytes is the active focus in the mechanism of liverfibrosis. Isolation and culture of the cells is of significant importance.METHODS According to Friedman's method we modified a more simple,convenient and economic method in isolation of lipocytes and Kupffer cellswith Ficoll - Hypaque for the first time.RESULTS Lipocytes and Kupffer cells were isolated sucessfully with highpurity and viability.CONCLUSION The technique for isolation of lipocytes and Kupffer cellsdescribed here is simple, reliable and economical.
