摘 要: | 目的 观察清肾颗粒对5/6肾切除肾纤维化模型大鼠肾组织P-选择素(P-selectin)/ P-选择素糖蛋白配体-1(P-selectin glycoprotein ligand 1,PSGL-1)表达的影响及其抗肾纤维化的作用机制。方法 将72只雄性SD大鼠随机分为正常组,模型组,清肾颗粒高、中、低剂量组,氯沙坦组,每组12只;除正常组外,剩余60只大鼠均按手术方法制备5/6肾切除肾纤维化模型,各组动物均从模型复制1周后开始灌胃给药,清肾颗粒各剂量组和氯沙坦组分别予以一定比例的清肾颗粒和氯沙坦水溶液灌胃,模型组和正常组均以等比例温水灌服。12周后处死大鼠,取残余肾组织用于检测P-selectin/PSGL-1蛋白及其mRNA的表达水平。结果 与正常组比较,模型组P-selectin、PSGL-1蛋白及其mRNA相对表达水平明显升高,差异均有统计学意义(P<0.05);与模型组比较,清肾颗粒各剂量组和氯沙坦组P-selectin、PSGL-1蛋白及其mRNA表达水平降低,差异均有统计学意义(P<0.05);与氯沙坦组比较,清肾颗粒高剂量组P-selectin、PSGL-1蛋白及其mRNA表达水平降低,差异均有统计学意义(P<0.05);清肾颗粒各组之间比较,清肾颗粒高剂量组P-selectin、PSGL-1蛋白及其mRNA表达水平最低,差异有统计学意义(P<0.05)。结论 清肾颗粒能够降低5/6肾切除大鼠肾组织P-selectin及PSGL-1的表达,这可能是其抗肾纤维化的机制之一。

Effect of Qingshen Granule on P-selectin and P-selectin Glycoprotein Ligand 1 in Rats with Renal Fibrosis |

Abstract: | Objective To investigate the effect of Qingshen Granule on the expression of P-selectin and P-selectin glycoprotein ligand 1 (PSGL-1) in renal tissue in 5/6 nephrectomized rats with renal fibrosis and its anti-renal fibrosis effect. Methods A total of 72 male Sprague-Dawley rats were randomly divided into normal group, model group, high-, middle-, and low-dose Qingshen Granule groups, and losartan group, with 12 rats in each group. All rats except those in the normal group were given surgical treatment to establish a 5/6 nephrectomized rat model of renal fibrosis, and the drugs were given by gavage since one week after modeling. The rats in the high-, middle-, and low-dose Qingshen Granule groups and the losartan group were given Qingshen Granule at a certain concentration or losartan (dissolved in water) by gavage, and those in the model group and the normal group were given an equal volume of warm water by gavage. The rats were sacrificed after 12 weeks and renal tissue was collected to measure the protein and mRNA expression of P-selectin and PSGL-1. Results Compared with the normal group, the model group had significant increases in the protein and mRNA expression of P-selectin and PSGL-1 (P<0.05). Compared with the model group, the high-, middle-, and low-dose Qingshen Granule groups and the losartan group had significant reductions in the protein and mRNA expression of P-selectin and PSGL-1 (P<0.05). Compared with the Losartan group, the high-dose Qingshen Granule group had significant reductions in the protein and mRNA expression of P-selectin and PSGL-1 (P<0.05). Among the three Qingshen Granule groups, the high-dose Qingshen Granule group had the lowest protein and mRNA expression of P-selectin and PSGL-1 (P<0.05). Conclusion Qingshen Granule can reduce the expression of P-selectin and PSGL-1 in renal tissue in 5/6 nephrectomized rats, which might be one of its anti-renal fibrosis mechanisms. |
