薯蓣皂苷元活性成分对心肌细胞损伤的保护作用及机制研究

摘    要:目的观察薯蓣皂苷元(MPD)活性成分对缺氧再复氧引起的心肌细胞损伤的保护作用,并研究其作用机制。方法采用建立缺氧再复氧引起心肌细胞损伤的模型,将细胞分为正常组、对照组和MPD组,正常组不进行缺氧再复氧和药物干预,对照组进行缺氧再复氧无MPD干预,MPD组进行缺氧再复氧和MPD干预,观察乳酸脱氢酶(LDH)和心肌细胞膜ATP酶。另外对正常心肌细胞经MPD的处理后,检测钠钙交换器(NCX)的mRNA表达量。结果与对照组比较,加入MPD(10μg/mL、50μg/mL)处理后,细胞的损伤程度减轻。经缺氧再复氧后MPD组的钠钾ATP酶和钙镁ATP酶的活性显著高于对照组。与对照组比较,NCX的mRNA表达量经MPD处理后降低(P0.05)。结论MPD可能通过维持心肌细胞膜钠泵和钙泵功能,共同影响心肌细胞钙离子跨膜转运,维持细胞内低钙离子的环境。
