摘    要:目的:建立适用于膜片组技术(Patchclamptechnigue,PCT)的大白鼠海马CA_4区锥体细胞(rathippocampalCApyramidalneuron,RHCPN)的急性分离方法,并测定该细胞的电生理学特性。方法:用7~21d大鼠,以链霉蛋白酶E酶解加吸管吹打法制备RHCPN,用全细胞PCT测定其电生理学特性。结果:分离的RHCPN易用于PCT,且具有正常的电生理学特性,并保存了主要的电压依赖性和受体门控性高于通道特性。结论:本法分离的RHCPN适用于PCT电生理学研究。

Abstract:To acutely isolate rat hippocampal CA1 pyramidal neurons (RHCPN) suitable forpatch clamp technique (PCT), and to measure their electrophysiological properties. Methods:RHCPNwere obtained by pronase E treatment and then by trituration with pipette from 7 to 21 day-old rats.PCT was used to study their electrophysiological properties. Results:The isolated RHCPN were easy torecord by PCT. Their electrical and chemical membrane preperties concerning some major voltage -de-pendent and receptor-gated ionic channels were well-preserved. ConcIusion: The RHCPN isolated bythis method is suitable for PCT electrophysiological study.
