脂肪酸合成酶在食管癌中的表达及临床意义

摘    要:
目的:研究脂肪酸合成酶在食管癌中的表达及其临床意义。疗法:通过免疫组织化学的方法对45例食管癌标本,6例不典型增生的食管粘膜和12例正常食管粘膜中的脂肪酸合成酶的表达进行检测;并与临床病理指标进行比较。结果:在45例食管癌中,FAS表达阴性的有6例,表达弱阳性的有4例,表达阳性的有23例,表达强阳性的有9例,表达极强阳性的有3例,阳性率78%;6例中度-重度不典型增生的食管粘膜中,1例表达阴性,2例表达弱阳性,2例表达阳性,1例表达强阳性,阳性率50%;而12例正常食管粘膜中,FAS表达阴性的有10例,弱阳性的有2例。食管癌中FAS的表达与临床病理指标无相关性。结论:脂肪酸合成酶在食管鳞型细胞的癌变过程中可能起重要的作用。脂肪酸合成酶在肿瘤细胞中选择性的分布可能对肿瘤治疗提供了新的靶位。

Abstract:
Objective: To explore the expression of fatty acid synthase (FAS) and its significance in esophageal carcinoma. Methods: We examined the immunohistochemical reactivity of FAS in 12 normal sophageal epithelium , 6 dysplastic esophageal lesions and 45 squmaous cell carcinomas. We analysed the correlation between FAS and various clinicopathological parameters in patients with esophageal carcinoma. Results: The FAS-postive cell was expressed 35 of 45 (78%) in esophageal carcinoma, 3 of 6 (50%) in dysplastic esophageal lesions. In contrast,normal esophageal epithelium did not express FAS. High expression of FAS in squmaous cell carcinomas did not correlate with clinicopathological features. Conclusion: The expression of FAS may pilay an important role in esophageal carcinoma . The FAS is expressed in esophageal carcinoma and dysplastic esophageal lesions suggesting that FAS may be upregulated continuously from esophageal squmaous cell carcinogenesis to established carcinoma. The selected distribution of FAS in tumor cells may provide a new way for targeting therapy.
