摘    要:目的观察消异方对子宫内膜异位症(EM)大鼠异位内膜生存素(survivin)、半胱氨酸蛋白酶-3(caspase-3)的影响,初步探讨消异方治疗子宫内膜异位症的作用机制。方法将80只雌性大鼠造模后随机分为空白对照组,假手术组,模型组,散结镇痛胶囊组,米非司酮组,消异方低、中、高剂量组共八组,灌胃28 d后,取正常子宫和异位内膜组织,用免疫组化的方法检测survivin和caspase-3的变化。结果消异方各组异位内膜的凋亡抑制基因survivin表达与模型组比较,差异均有统计学意义(P〈0.05或P〈0.01);caspase-3表达与模型组比较,差异亦有统计学意义(P〈0.05)。结论消异方通过激活细胞中的caspase酶原活性及降低survivin在EM中的高表达,影响异位内膜细胞的凋亡,抑制异位内膜的增长。

Abstract:Objective To observe the impacts of xiaoyi formula on the expression of survivin and caspase-3 for the rats with endometriosis(EM)and explore initially the mechanism of EM so as to provide the important evidence for clinical practice.Methods 80 female rats after modeling were divided randomly into a blank control group,a sham-operation group,a model group,a sanjie zhentong capsules group,a mifepristone group,a low-dose xiaoyi formula group,a middle-dose xiaoyi formula group and a high-dose xiaoyi formula group.Totally,8 groups were included.In 28 days of gastric infusion,the normal uterine and ectopic endometrial tissues were collected.Immunohistochemical assay was adopted to detect the changes in survivin and caspase-3 expressions.Results The apoptosis gene survivin expression in ectopic endometrial tissue in each xiaoyi formula group was significantly different in statistics as compared with model group(P<0.05 or P<0.01).The expression of caspase-3 was different statistically as compared with model group(P<0.05).Conclusion Through activating caspase plasminogen activity and reducing survivin high-expression in the cells,xiaoyi formula affects the apoptosis of ectopic endometrial cells and inhibits ectopic endometrial increase.
