摘    要:
目的 下调T24膀胱癌细胞株中Raf1的表达,观察癌细胞株的细胞凋亡情况.方法 使用SiRNA干扰方法阻断Raf1在T24膀胱癌细胞株中的表达,随后用流式细胞仪观察细胞凋亡情况,使用westem-blot法确认细胞凋亡途径.结果 使用SiRNA干扰后Raf1表达显著降低,随之凋亡细胞数量明显增高,western-blot法确认Raf1表达降低后Caspase-3和PARP蛋白被激活.结论 Raf1表达抑制促使T24细胞凋亡,其凋亡通路是通过Caspase-3和PARP途径,此研究可能对膀胱癌的治疗提供新思路.

Reduction of Raf1 expression to promote the apoptosis in T24 bladder cancer cell line
Abstract:
Objective Down-regulation the Raf1 expression can induce the apoptosis of T24 bladder cancer cell line.Methods We used SiRNA interference technology to block the Raf1 expression in T24 bladder cancer cell line.Subsequently we tested the apoptotic cells changes by flow cytometry assay,and confirmed the apoptosis pathway by the western blot assay.Results After the SiRNA interference,the Raf1 expression decreased apparently.Subsequently,the apoptotic cells increased remarkably.And the western-blot assay of Caspase-3 and PARP showed that both two proteins were activated by the reduction of Raf1.Conclusion Reduction of the Raf1 expression can make T24 cells apoptosis.And the apoptotic pathway is through the Caspase-3-PARP mechanism.This study provides a new idea to handle with bladder cancer.
