摘    要:
目的研究低氧对体外培养胃上皮细胞株GES-1中MHC-Ⅰ类链相关蛋白A(MICA)基因表达的影响。方法建立GES-1细胞体外低氧培养模型,免疫细胞化学技术分别检测MICA在缺氧处理0、24、72 h后的蛋白表达。结果免疫细胞化学证实MICA蛋白的表达随着缺氧时间的增加呈时间效应关系,且表达量随缺氧时间而升高(P〈0.05)。结论低氧可以显著增加胃上皮细胞株GES-1中MICA的表达。

Abstract:
Objective To investigate the effect of hypoxia on major histocompatibility complex class Ⅰ chainrelated protein A(MICA) expression in gastric epithelial cell strain-1(GES-1) and its mechanism.Methods Hypoxia culture model in vitro was established to test the expression of MICA in GES-1 cell line.MICA expression was tested in protein level by using immunocytochemistry analysis after cultured in hypoxia circumstance for 0,24,and 72 hours,respectively.Results MICA expressions increased after treatment of hypoxia with time-effect relationship(P〈0.05).Conclusion MICA expression in GES-1 can be induced remarkably by hypoxia.
