c－myc癌基因产物P62的表达与膀胱癌生物学行为关系

应用胶体金免疫电镜技术，检测９例膀胱癌标本中Ｐ６２的定位，发现Ｐ６２定位于细胞核。采用ＡＢＣ法免疫组化技术，检测１３例正常膀胱组织、１４例膀胱慢性炎性增生组织和５１例膀胱移行细胞癌标本中Ｐ６２的表达，并用显微分光光度计测得每例膀胱癌标本Ｐ６２的表达强度，与临床资料对比，结果发现正常膀胱组织和膀胱慢性炎性增生组织标本Ｐ６２染色阴性，５１例膀胱移行细胞癌标本中３４例Ｐ６２染色阳性，Ｐ６２表达的强度与膀胱癌的病理分级、临床分期呈正相关，与预后呈负相关。

Studies on the relationship between expression of c-myc oncogene product P62 and bladder tumor biologic behaviors

The colloid-gold immunoelectron microscopy was used to detect the expression and distribution of P62 colloid-gold granules were detected in the nucleus of bladder tumor cells under electronmicroscopy.P62 McAb was used to detect by immunohistochemical assay the expression of P62 in the paraffin embedded specimens from l3 normal bladder tissues,14 chronic inflammatory bladder hyperplasia tissues and 5l bladder tumor tissues.Microspectrophotometer was used to detect the density of P62 of the specimens,and the densities of P62 were compared with the pathological grading,the clinical staging and the prognosis of the patients.The immunohistochemistry assay showed that P62 was neither found in normal bladder tissues nor in chronic inflammatory hyperplastic tissues,but were detected in some of bladder tumor tissues as 34 of 5l bladder tumor tissues showed positive expression of P62.Compared the microspectrophotometer assay data and the clinical follow up of the patients also showed that the density of P62 expression was positively related to the pathological grading and the clinical staging.The expression of P62 in cases of recurrence and death were more remarkable.
