摘    要:作用设计合成的两对引物建立了巢式PCR-EB染色法检测B19DNA新技术,扩增产物为104bp片段,敏感性达0.007fg,是一般PCR的100倍,是一种特异、敏感、简便、快速诊断B19感染的新方法。用该方法检测了57例自然流产和死胎的胎儿组织,17例阳性,其中自然流产11/42(265),死胎6/15(40%),证实了国内胎儿有B19病毒感染,和自然流产、死胎可能相关,并对PCR实验中防污染措

Detection of human parvovirus B19 infections in fetal tissues by nest-ed PCR
Abstract:A new nested PCR-EB staining assay technique to detect B19 DNA was established by using two pairs of primers system. The amplified Products are 104 bp DNA fragments and its sensitivity is 0.007 fg,100 times more sensitive than the single PCR. This method is specific,sensitive,simple and rapid in diagnos- ing B19 infections.By this method,57 fetal tissues of spontaneous abortions and still births were detected. Among which 17 were positive.In these positive samples,spontaneous abortions were 11/42(26%)and still births were 6/15(40%).These results verify that there are B19 infections in fetus in China and that there may be possible relativity among spontaneous abortion,still birth and B19 infection. Furthermore, measures of avoiding contamination in PCR tests were also discussed in this study.
