银杏叶提取物对大鼠脑缺血再灌注后Bcl-2、Bax蛋白和Caspase-3表达的影响

摘    要:目的探讨银杏叶提取物(ginkgo biloba extract,GBE)对脑缺血再灌注后Bcl-2、Bax蛋白和半胱氨酸蛋白酶-3(Caspase-3)表达的影响。方法将SD大鼠随机分为假手术组(A组)、缺血再灌注组(B组)和GBE治疗组(C组),建立大鼠大脑中动脉缺血1.5 h再灌注24 h的模型;C组于缺血前30 min及缺血再灌注后1 h腹腔内注射银杏叶提取物;采用免疫组化SP法检测脑缺血再灌注后Bcl-2、Bax蛋白的表达,采用四肽酶底物法检测Caspase-3活性水平。结果 GBE治疗组大鼠Bcl-2阳性细胞表达数目均高于缺血再灌注组,Bax少于缺血再灌注组(P〈0.05);假手术组未检测到Caspase-3活性升高,缺血再灌注组Caspase-3活性明显高于GBE组,有统计学意义(P〈0.05)。结论 GBE可通过上调Bcl-2蛋白表达而减少Caspase-3活性水平,下调Bax蛋白表达,来降低脑缺血再灌注后大鼠神经细胞凋亡。
