实时荧光PCR检测胃癌石蜡组织中Her-2表达的研究

目的探索胃癌石蜡块组织中Her-2的表达状态及其检测新方法。方法统计2009～2011年行免疫组化检测其Her-2蛋白表达情况的71例胃癌组织蜡块的病理数据,并用实时荧光聚合酶链反应(PCR)检测其样本中的Her-2基因的扩增情况,将两种检测方法的结果进行比较。结果胃癌组织中Her-2的过表达率:免疫组化法为22.5%,荧光PCR法为19.7%,Her-2的过表达与胃癌的部位、有无淋巴结转移及TNM分期比较有显著的相关性(P<0.05),与年龄、性别、分化程度、大体分型、浸润深度比较无显著相关性(P>0.05)。两种方法的检测结果无明显差异(P>0.05)。结论Her-2在胃癌组织表达增高与胃癌的发生、发展密切相关。实时荧光PCR简便、客观、高效,可望成为胃癌石蜡组织中除免疫组化、荧光原位杂交外检测Her-2变化的一种备选方案。

Research of Real-time Fluorescent PCR Testing the Expression of Her-2 in Paraffin-embedded Tissues of Gastric Cancer

Objective To explore the expression status of Her-2 in paraffin blocks of the gastric cancerand the new test methods.Methods Pathological data of the expression of Her-2 protein in 71 cases of gastric carcinoma tested by immunohistochemistry in 2009-2011 were reviewed,and the expression of Her-2 gene of the sample were tested by real-time PCR,the results of the two testing methods were compared.Results Her-2 over-expression of gastric cancer rate:22.5% by immunohistochemical method,19.7% by fluorescent PCR method.Her-2 over-expression had significant correlation with the site of gastric cancer,lymph node metastasis and TNM stage(P<0.05),and had no significant correlation with age,gender,degree of differentiation,gross type,and depth of invasion(P>0.05).The result of the two methods of examination showed no significant difference(P>0.05).Conclusion There is close correlation between Her-2 over-expression in gastric cancer genesis and development.Real-time PCR is simple,objective and efficient,and is expected to be another option of testing the expression of Her-2 in paraffin-embedded tissues of gastric cancer in addition to immunohistochemistry and FISH test.
