微管蛋白酪氨酸连接酶样-1在小鼠脊髓中表达差异的研究

摘    要:
目的:探讨微管蛋白酪氨酸连接酶样-1(Ttll1)在脊髓发育中的作用。方法:选用孕18天胎鼠及出生后1天、5天、10天、20天、40天的A/J(A)和C57BL/6J(B6)小鼠,以实时荧光定量PCR方法测定各发育阶段Ttll1在脊髓中的表达。结果:2种品系小鼠不同发育阶段Ttll1的表达有明显差异,A/J小鼠在出生后1天表达最强,C57BL/6J小鼠在胚胎18天表达最强且峰值显著高于A/J的峰值。提示Ttll1的表达与脊髓的发育及与两种小鼠脊髓发育的差异有关。结论:Ttll1基因与脊髓发育相关,其机制可能是通过表达强度调节脊髓的发育。

Abstract:
Objective: To explore the role of tubulin tyrosine ligase-like 1(Ttll1) in the development of spinal cord in mice. Methods: By means of real time-polymerase chain reaction(PCR), expressions of Ttll1 in spinal cord in two inbred strains of mouse, A/J(A) and C57BL/6J(B6), were detected at different developmental phases, E18,P1d,P5d, P10d, P20d and P40d. Results: Expressions of Ttll1 in two inbred strains showed obvious difference at different developmental phases, the peak of expression in A/J mouse was at P1d, while C57BL/6J was at E18, and the level was higher than A/J markedly. These results indicated that expression of Ttll1 gene related to the development of spinal cord and the difference of the expression may be one of mechanism modifying development of spinal cord between the two strains. Conclusion: Ttll1 gene is related to the development of spinal cord ,and it probably regulates the development of spinal cord by the intention of expression.
