趋化性细胞因子ENA-78与子宫内膜异位症的关系

目的:检测子宫内膜异位症(EM)患者腹腔液中趋化性细胞因子ENA-78水平及ENA-78在异位组织中的表达,探讨趋化性细胞因子ENA-78在EM发病机制中的作用。方法:用酶联免疫吸附试验测定EM患者(实验组)和子宫肌瘤患者(对照组)腹腔液中ENA-78水平;同时采用免疫组化方法检测异位内膜组织与在位内膜组织ENA-78的蛋白表达。结果:①实验组腹腔液ENA-78水平显著高于对照组(P<0.05),并随着疾病的严重程度而增高(P<0.05);②ENA-78表达于异位腺体细胞、间质细胞及巨噬细胞的胞浆中,表达强度随着疾病的严重程度而增强(P<0.05),不同月经周期中ENA-78表达的差异无统计学意义(P>0.05);③EM患者在位内膜ENA-78表达较对照组强(P<0.05)。结论:ENA-78是一种重要的炎性介质,具有趋化并活化炎性细胞活性及促进异位内膜病灶血管增生,在EM的发生和发展各个环节均发挥作用,并随着疾病进展表达增强,是反映EM侵袭生长的生物学指标。

Relationship between chemotactic cytokine ENA-78 and endometriosis

Objective:To detect the expression levels of chemotactic cytokine epithelial neutrophil-activating peptide-78 (ENA-78) in peritoneal fluid and ectopic tissues of patients with endometriosis,explore the role of ENA-78 in pathogenesis of endometriosis.Methods:The levels of ENA-78 in peritoneal fluid of patients with endometriosis (study group) and patients with hysteromyoma (control group) were detected by enzyme-linked immunosorbent assay (ELISA).The expression levels of ENA-78 protein in ectopic and eutopic tissues were measured by immunohistochemical technique.Results:①The level of ENA-78 in peritoneal fluid in study group was significantly higher than that in control group (P<0.05),increasing with the aggravation of disease (P<0.05).②ENA-78 located in cytoplasm of ectopic glandular cells,stromal cells and macrophages,expression levels increased with the aggravation of disease (P<0.05),there was no significant difference in ENA-78 expression between different menstrual cycles (P<0.05).③The expression of ENA-78 in eutopic tissues of study group was higher than that of control group (P<0.05).Conclusion:ENA-78 is an important inflammatory media,which can activate the activity of inflammatory cells,promote the angiogenesis of ectopic endometrial tissues and play an important role in the occurrence and development of endometriosis,increase with the progression of endometriosis.At the same time,it is a biological indicator of invasion of endometriosis.
