摘    要:目的研究碱性、中性两种条件下CNBr活化Sepharose 4B偶联欧蔓陀罗凝集素(DSA)的效果。方法在两种偶联条件下,通过改变CNBr用量、活化时间以及温度等条件,观察DSA的偶联率指标。结果碱性条件下,活化15 min即使增加CNBr用量至300 mg/ml凝胶,固定化凝集素偶联率没有提高,DSA偶联率较低,在29.1%~50.9%之间;中性条件下,CNBr用量仅为碱性条件下的20%,活化温度控制在-10~-20℃可能获得较好的活化效果,DSA偶联率在78.8%~80.9%之间。结论中性条件下CNBr活化Sepharose 4B偶联DSA,实验易于控制,DSA偶联率比较稳定,实验重复性好于碱性条件下的结果,且可获较高偶联率的DSA-Sepharose 4B。

Abstract:PurposeTo study the effect of coupling DSA with CNBr-Activated Sepharose 4B under the alkaline and neutral condition.MethodsIt was observed the percentage of coupling DSA with CNBr-activated Sepharose 4B under variable conditions such as duration of CNBr activation,experiment temperature as well as dosages of CNBr udner different pH condition.ResultsUnder alkaline condition,the percentage of coupling DSA with CNBr-activated Sepharose 4B is between 27.0% and 50.9%, at the same time,under neutral condition,the percentage of coupling DSA with CNBr-activated Sepharose 4B is between 78.8% and 80.9%.ConclusionUnder neutral condition,the experiment is easy to be controlled,the percentage of coupling DSA with CNBr-activated Sepharose 4B is much higher than that under alkaline condition.
