非肥胖型糖尿病小鼠自发性涎腺炎的发生与发展

摘    要:目的 观察雌性非肥胖型糖尿病(NOD)小鼠自发性涎腺炎的发生发展过程。方法 选择5、10、15、20周龄雌性NOD小鼠各6只。测定刺激全唾液流率(STFR)、施墨试验、唾液总蛋白、常规HE切片及电镜超微结构观察涎腺组织的改变。以BALB/c小鼠作对照。结果 10、15、20周NOD小鼠STFR、唾液总蛋白均明显低于对照组(P〈0.05),相应的下颌下腺分泌颗粒减少。10周雌性NOD小鼠涎腺炎发病率为4/6,15、20周均为6/6。淋巴细胞浸润主要见于下颌下腺,舌下腺极少,腮腺未见。10周时NOD小鼠已有淋巴细胞浸润灶形成。15周显著增多而且面积增加。STFR与淋巴细胞浸润灶数呈负相关。结论 雌性NOD小鼠5~10周淋巴细胞开始浸润下颌下腺,刺激唾液和蛋白分泌降低。不同腺体受累情况不同。

Abstract:Objective To observe the process of sialoadenitis in non-obese diabetic(NOD)mice.Methods Six female NOD mice at age of 5,10,15 and 20 weeks respectively were matched with BALB/c mice.Total saliva flow rate(STFR),Schirmer test,total saliva protein,number of lymphocytes foci(NLF)were measured from both NOD and BALB/c mice.The ultrastructure of major salivary glands was observed.Results Sialoadenitis was not evidenced in 5-week NOD.However,the incidence of sialoadenitis was 4/6 in 10-week NOD and 6/6 in both 15 and 20-week groups.Lymphocytes foci were seen in the salivary glands of 10-week NOD and increased obviously in the salivary glands of 15-week NOD and further of 20-week NOD.STFR was decreased obviously after 10-week NOD.NLF showed negative relation with STFR.Lymphocytes infiltration was mainly found in submandibular glands,seldom in sublingual glands,and none in parotid glands.Conclusion Sialoadenitis occurred in female NOD mice aged 10 weeks and aggravated from the age of 15 weeks,which paralleled with the changes of STFR and NFL in submandibular glands.
