地塞米松诱导幼龄大鼠胸腺细胞凋亡模型

摘    要:
目的:建立一个比较实用且成型时间短的地塞米松诱导大鼠胸腺细胞凋亡模型。方法:给幼龄大鼠(4周 ̄5周)腹腔注射地塞米松(0.02g/kg)采用形态学(光镜和电镜)、DNA琼脂糖凝胶电泳、流式细胞光度分析等方法研究3、6、9、15、24h等不同注射时间胸腺细胞凋亡变化。结果:在15h内,地塞米松诱导的大鼠胸腺细胞凋亡百分率随时间延长,凋亡发生率从6.2%逐渐增至58.5%,注射地塞米松15h,在光镜和

Abstract:
Objective:To develop an apoptotic model of thymocytes in infancy rats with an intraperitoneal injection of dexamethasone.Methods:Three,six,nine,15 and 24 hours after introperitoneal injection of dexamethasone(0 02 g/kg) in infancy rats,the thymocyte apoptosis was studied and analysed by using morphological method,DNA agarose gel electrophoresis and flow cytometry.Results:During 15 hours,the apoptotic rate of rat thymocytes induced by dexamethasone increased with time (6 2% 58 8%).The optimum time was 15 hours after injection for the inducing,in which typical apoptotic morphological changes,ladder in DNA agarose gel electrophoresis,and apoptotic peak in flow cytometric analysis were observed.Conclusion:The thymocyte apoptosis was successfully induced by introperitoneal injection of dexamethasone(0 02 g/kg),which provides a valuable reference for the mechanism study of thymocyte apoptosis and clinical practice.
