抑癌p53基因在星形细胞瘤中阳性表达的意义

摘    要:目的研究p53在星形细胞瘤发生、增殖过程中的作用及临床意义。方法采用免疫组织化学法(S-P法)检测62例资料完整的星形细胞瘤及10例正常脑组织的p53表达,分析p53蛋白在星形细胞瘤及正常脑组织中的表达情况。结果正常脑组织中未见p53蛋白阳性表达,62例星形细胞瘤中共有26例肿瘤p53蛋白表达呈阳性,二者差异有显著性(χ2=4.872,P<0.05),其中在Ⅰ级星形细胞瘤中阳性率为8.33%(1/12),Ⅱ级星形细胞瘤中阳性率为21.05%(4/19),Ⅲ级间变性星形细胞瘤中阳性率为60%(9/15),Ⅳ级胶质母细胞瘤中阳性率为75%(12/16)。结论星形细胞瘤中p53蛋白的表达与肿瘤的级别相关,分化程度越低,p53蛋白阳性表达率越高,检测p53有助于临床判断预后。

Abstract:Objective To investigate the role of p53 during the proccess of the astrocytoma occurrence and cell proliferation,so as the clinical significance. Methods The positive expression of p53 protein were detected in the specimens of 62 cases of astrocytoma by S-P immunological technique,aside with 10 cases of normal brain tissue. Then the expression of p53 protein in these tissues were analysed. Results There were 26 cases positive expression of p53 protein in 62 astrocytoma specimens and no positive expression of p53 protein in the normal brain tissue. It showed a significant diversity between the two groups(x^2 = 4. 872, P d0.05). The positive percentage was 8.33 % (1/12) in grade Ⅰ astrocytomas,21.05%(4/19) in grade Ⅱ astrocytomas,60% (9/15) in grade Ⅲ anaplastic astrocytomas(AA) and 75%(12/16) in grade Ⅳ glioblastoma multiforme(GBM). Conclusion The results reveal a correlation between p53 protein expression and histopathological typing and grading in astrocytomas. The degree of cell differentiation is lower, the ratio and intensity of p53 expression is obviously higher, which can help to evalue prognosis.
