| 摘 要： | 目的：研究青蒿琥酯抗肝癌的作用机理。方法：检测拓扑异构酶活性，甲基绿－派络宁染色检测凋亡细胞，免疫组化方法观察Bax、Bcl-2的表达。结果：经青蒿琥酯处理的SMMC－7721细胞拓扑异构酶活性增强，青蒿琥酯处理后，凋亡细胞明显增加，免疫组化显示，青蒿琥酯组肿瘤标本Bax蛋白阳性细胞数增多，Bcl-2蛋白阳性细胞数减少。结论：青蒿琥酯抗肝癌作用机理可能为上调Bax基因，下调Bcl-2基因， 诱导癌细胞凋亡，同时影响拓扑异构酶活性。

| Abstract: | Objective:To study the mechanism of Artesunate inhibiting hepatocarcinoma.Methods:We assay the activity of the topoisomerase of hepatocarcinoma cells.The methyl green prynoin staining and the immunohistochemical staining was used to detect apoptotic cells and the tumor associated protein.Results:After the administration of Artesunate,the apopotic cells increased significantly,the immunohistochemical staining showed that Artesunate downregulates the expression of Bcl 2 genes and upregulates the expression of Bax genes.Conclusion:The mechanism of Artesunate may partly be inducing hepatocarcinoma cells to undergo apoptosis.Artesunate also interferes the activity of topoisomerase. |
