关节腔内注射医用臭氧对兔膝骨性关节炎的影响

摘    要:目的观察膝关节腔内注射医用臭氧对兔膝骨性关节炎的影响,研究不同浓度医用臭氧对骨性关节炎的治疗效果。方法将健康家兔32只随机分为A组(空气组)、B组(25μg/ml医用臭氧组)、C组(40μg/ml医用臭氧组)、D组(80μg/ml医用臭氧组),每组兔右膝关节均使用Hulth法复制出膝骨性关节炎(OA)模型,造模成功后,每组每周1次给予膝关节腔内注射医用臭氧,每次2ml,共4周。末次注射后3d处死实验兔,取股骨内侧髁附近软骨组织行病理学观察及Mankin评分;取关节液行NO检测;分别在造模前、造模成功时和处死前测定兔膝关节活动度。结果各组造模成功时右膝关节活动度较造模前降低,处死前B、组C组和D组右膝关节活动度较造模成功时升高,B组处死前即刻右膝关节活动度较C组和D组明显升高(P〈0.05);各组右膝关节软骨病理检查均出现关节软骨损害改变,B组关节软骨退变程度轻,Mankin评分较左膝差异无统计学意义(P〉0.05),C组和D组关节软骨退变明显,Mankin评分较左膝差异有统计学意义(P〈0.05);各组右膝关节关节液中NO含量升高,B组关节液中NO含量相对于C组和D组降低程度最大。结论三种浓度的医用臭氧均能够减轻骨性关节炎的炎症反应,而40μg/ml和80μg/ml的医用臭氧会导致关节软骨组织结构破坏,臭氧浓度越高损害越重。

Effect of intra-articular injection of medical ozone in rabbits of knee osteoarthritis

Abstract:Objective To observe the impact of articular injection of medical ozone knee osteoarthritis in rabbits and discuss the treatment of different concentrations of medical ozone on osteoarthritis.Methods 32 rabbits were randomly divided into 4 groups of A (air group),B (25 μg/ml medical ozone group),C (40 μg /ml group medical ozone),D (80 μg /ml medical ozone group),each right knee joint were modeled,then give the knee articular injection of 2 ml medical ozone once weekly,a total of 4 weeks.3 days after the last injection the rabbits were killed,taken the cartilage near the medial femoral condyle for pathological observation and Mankin score;take joint fluid for NO detection;measure the degree of knee before modeling,modeling successful and before execute.Results 1.When modeling successful each group right knee degrees was lower than before modeling.Before executed B,C,D group right knee degrees was higher than when modeling successful.Before executed B group right knee degrees was significantly higher than C group and D group (P<0.05).2.Articular cartilage pathology damage was found in right knee of each group,articular cartilage of B group was mild degeneration.Mankin score of right knee articular cartilage was similar as that in the left knee;the articular cartilage had significant degeneration in C group and D group;There was significant difference of mankin score of articular cartilage between their right knee and left knee (P<0.05).3.NO level of the joint fluid of right knee rised in each group,the NO level of B group reduce the maximum degree in relative to C group and D group.Conclusion Three levels of medical ozone were able to reduce the inflammation of osteoarthritis.40μg/ml and 80μg/ml of medical ozone can cause structural damage to articular cartilage,and the damage was increased with increasing concentrations of medical ozone.
