ANIT诱发大鼠肝内胆汁淤积的肝肾功能动态变化研究

摘    要:目的观察ANIT诱发大鼠胆汁淤积形成过程中模型肝、肾功能动态变化特点。方法一次性给予100mg/kg体重ANIT复制大鼠肝内胆汁淤积模型;于造模后24h、48h、72h、96h分4批杀鼠取材,观测大鼠一般状况,肝、肾功能。结果与赋形剂组比较,模型组大鼠的体重都有不同程度的降低;肝重,肝指数呈增高的动态变化;脾重,脾指数先增高后降低的动态变化;血清AlB含量在24、48、72h维持在较低水平,到造模96h又恢复至接近正常水平;GLO、TP、CHO含量造模后无明显变化;TBil、TBA含量,ALT、AST活性在造模24h和72时呈明显增高的动态变化(P〈0.01),在造模48h和96h时接近正常水平;ALP、GGT在造模后有显著增高(P〈0.01),96h时数值降低;LDH活性在造模后显著下降(P〈0.01),96h时升高至正常水平;BUN含量在造模后24h时有所下降,随后逐渐增高接近正常水平;CRE含量在48h,72h时显著增高(P〈0.01),96h时接近恢复正常。结论①ANIT所诱发的肝内胆汁淤积大鼠模型病变表现为急性肝内胆汁淤积,肝细胞损伤。②血清TBil、TBA含量,ALT、AST活性的特殊变化提示有2次致损可能。

Abstract:Objective To observe the change characteristics of the liver and renal function during the course of the model liver tissue of rat cholestasis induced by ANIT.Methods ANIT (100mg/kg) was administered all at once to establish the cholestasis model in rat liver.The rats were killed in four batches at 24h,48h,72h,and 96h after establishing the model for getting the requisite tissue.We observed the rat physical signs and the liver and renal function.Results Compared with the excipient group,there is rat weight loss in vary degree in the model group;the liver weight and index are raising;the spleen weight and index are raising early and then depressed;the serum ALB content maintain the lower level when 24h,48h and 72h,and come back to the normal level when 96h;there is no remarkable change in GLO,TP and CHO content after establishing the model;there is significant increasing change in TBil content,TBA content,and in ALT and AST activity(P0.01),which maintain the normal level when 48h and 96h;ALP and GGT are significantly increasing after establishing the model(P0.01),but decreasing when 96h;LDH activity is significantly decreasing(P0.01),but come back to the normal level when 96h;BUN content is decreasing when 24h,later come back to the normal level gradually;CRE content is significantly increasing when 48h and 72h(P0.01),and come back to the normal level when 96h.Conclusion ①the pathological change of choleasstis in rat liver induced by ANIT are characterized by the cholestasis in liver and the liver cell damage;②the unique change of the TBil,TBA,ALT and AST indicates the possibility of the twice damages.
