RP-HPLC示差折光检测法测定复方知母胶囊中菝葜皂苷元的含量

 目的 采用反相高效液相色谱示差折光检测法建立复方知母胶囊中菝葜皂苷元的含量测定方法。方法 色谱柱:Hy-persil ODS(4.6 mm×150 mm,5μm)不锈钢柱,流动相:纯甲醇,流速:0.9 mL·min^-1,检测器:RID2410,柱温:30℃,进样量:20μL。结果 被测定峰与其它色谱峰可达到基线分离,线性范围1.0～7.0μg,相关系数r=0.999 8,平均回收率98.66％,(RSD=2.36％,n=5)。结论 本法简便,准确,灵敏度高,可用于本制剂的含量测定和质量控制。

Determination of sarsasapogenin in compound Zhimu capsules by HPLC-RI

OBJECTIVE To establish a method for the determination of sarsasapogenin in compound Zhimu capsules by RP-HPLC-RI. METHODS 5 um Hypersil ODS column (4.6 mm×150 mm) was used and the mobile phase was methanol with the flow rate of 0.9 mL·min^-1. It was detected by RID 2410 detector with column temperature at 30℃. The injection volume was 20 uL. RESULTS Sarsasapogenin and other components were well separated. The standard curve was linear over the range of 1.0～7.0μg with the correlation coefficient 0.999 8, the average recovery was 98.66% (RSD = 2.36% ,n=5). CONCLUSION The method was simple, accurate and sensitive for the determination of Sarsasapogenin and the quality control for compound Zhimu capsules.
