THP1单核细胞分化过程中MMP9表达量的变化

目的研究单核细胞在向巨噬细胞、泡沫细胞分化过程中基质金属蛋白酶-9(MMP-9)基因和蛋白表达的变化。方法体外诱导人单核细胞系(THP-1)细胞株分化为巨噬细胞和泡沫细胞,采用荧光定量聚合酶链式反应和Western blotting分别检测单核细胞向巨噬细胞、泡沫细胞分化过程中MMP-9基因和蛋白表达的变化。结果THP-1单核细胞向巨噬细胞、泡沫细胞分化过程中MMP-9 mRNA表达量分别增加了2.03倍和3.36倍(P<0.05,P<0.01);蛋白表达分别增加了5.86倍和3.68倍(P<0.01,P<0.05)。巨噬细胞和泡沫细胞中的MMP-9 mRNA水平无明显差异(P>0.05),但泡沫细胞中蛋白表达量较巨噬细胞有显著减少(P<0.05)。结论经体外诱导分化成的巨噬细胞和泡沫细胞中MMP-9 mRNA、蛋白水平与单核细胞比较呈显著增加,但单核细胞分化为泡沫细胞后其蛋白表达量呈显著下降。

Expression of matrix metalloproteinases 9 during THP 1 monocytes differentiation of into macrophages and foam cells

Objective To observe expression of MMP-9 mRNA and protein during THP-1 monocytes was induced into macrophages and foam cells. Methods THP-1 monocytes were differentiated to macrophages in vitro and then were induced into foam cells by culturing with oxLDL. MMP-9 mRNA and protein expression were determined by quantitative RT-PCR and Western blot,respectively. Results The expression of MMP-9 mRNA increased about 2.03 fold(P<0.05) and 3.36 fold(P<0.01) during THP-1 monocytes inducing into macrophages and foam cells.At the same time,the MMP-9 protein level also increased about 5.86 fold(P<0.01) and 3.68 fold(P<0.05),respectively.However,expressions of MMP-9 mRNA had no significant difference between macrophages and foam cells(P>0.05).MMP-9 protein expression was lower in foam cells than in macrophages(P<0.05). Conclusion MMP-9 mRNA and protein expression were both significantly increased during THP-1 monocytes inducing into macrophages and foam cells.But MMP-9 protein expression decreased more in foam cells.
