不同成熟时期山羊卵母细胞冷冻保存研究

摘    要:目的比较山羊卵母细胞在不同成熟时期解冻后体外成熟、体外受精及胚胎发育能力。方法收集山羊生殖泡(GV)期未成熟卵和成熟卵,分5组行开放式拉细麦管(OPS)法玻璃化冷冻,Ⅰ组:直接冷冻GV期卵;Ⅱ组:GV期卵在体外培养(IVC)8h后冷冻;Ⅲ组:GV期卵在IVC16h后冷冻;Ⅳ组:GV期卵体外培养成熟(IVM)后冷冻;Ⅴ组:直接冷冻体内成熟卵。解冻后比较各组卵受精率、卵裂率及8细胞胚胎形成率。结果Ⅰ组(GV期卵)冷冻解冻后存活率达56.8%,显著高于Ⅱ组(33.3%)、Ⅲ组(31.6%)、Ⅳ组(28.9%)及Ⅴ组(30.4%),差异有高度统计学意义(均P〈0.01)。各组体外受精率、卵裂率及8细胞胚胎形成率比较,差异无统计学意义(P〉0.05)。结论采用OPS法玻璃化冷冻山羊GV期卵较冷冻其他成熟时期卵效果好。

Study on Cryopreservation of Goat Oocytes at Different Developmental Stages

Abstract:Objective To examine maturation(IVM) in vitro, fertilization(IVF) in vitro and embryo developmental capacity of thawed oocytes frozen in the different developmental stages. Methods The germinal vesicle (GV) and mature goat oocytes were randomly divided into five groups for vitrification using open pulled straw(OPS). Group I : frozen at GV stage. Group Ⅱ : frozen after culture(IVC) in vitro 8 h. Group m: frozen after IVC 16 h. Group IV: frozen after IVM. Group .V: frozen oocytes cultured in vitro. Then the thawed oocytes were cultured for observing IVF, cleavage and 8-cell rates. Results The survival rate of oocytes frozen in the GV stage was 56.8%, which was significantly higher than that in Group Ⅱ (33.3%) statistically, Group m(31.6%), Group Ⅳ(28.9%), Group Ⅴ (30.4%) (all P〈0.01). There was no significant difference statistically in the rate of IVF, cleavage and 8-cells among three groups(P〉 0.05). Conclusion The goat oocytes at GV stage can be more effectively vitrified using OPS than that in the other developmental stages.
