摘    要:目的研究Necrostatin-1(Nec-1)对帕金森病(Parkinson's disease,PD)细胞模型线粒体失能的影响,探讨Nec-1在帕金森病发病机制中的作用。方法经不同浓度Nec-1预处理的PC12细胞加入6-OHDA建立多巴胺能神经元损伤模型。用4-甲基偶氮唑蓝(MTT)法检测PC12细胞活力,JC-1试剂盒检测线粒体膜电位,免疫印迹法检测Cathepsin B与Bcl-2蛋白表达。结果 100μmol/L的6-OHDA处理PC12细胞24h,细胞活力较正常对照组明显降低(P<0.05);与模型组比较,5、10、20、30、60μmol/L的Nec-1可明显减轻6-OHDA的毒性,90μmol/L的Nec-1可加重6-OHDA的毒性。Nec-1可阻抑线粒体膜电位下降。与正常对照组相比,模型组Cathepsin B表达明显增加而Bcl-2的表达显著降低(P<0.05)。与模型组相比,Nec-1与UTI可降低Cathepsin B表达并使Bcl-2的表达逐渐增加(P<0.05)。结论 Nec-1在一定的浓度范围内可提高PC12细胞存活力,通过稳定线粒体膜电位、抑制Cathepsin B的表达、上...
