柴胡疏肝散对抑郁症大鼠海马神经递质含量的影响

摘    要:目的探讨柴胡疏肝散对抑郁症大鼠海马神经递质含量的影响。方法将60只sD大鼠随机分为空白组、模型组、氟西汀组、柴胡疏肝散中剂量组和高剂量组,每组12只。除空白组外其余4组大鼠连续14天腹腔注射利血平注射液造模。造模后氟西汀组和柴胡疏肝散中、高剂量组每天分别给予9mL/kg氟西汀、5g/kg和10g/kg柴胡疏肝散灌胃。给药开始后第0、4、7、10、14天测量体重,第0、7、14天计算Open—Field行为得分,第15天处死动物,采用ELISA法测定海马组织单胺类神经递质5-羟色胺(5-HT)、去甲。肾上腺素(NE)、多巴胺(DA)的含量。结果与模型组比较,柴胡疏肝散高剂量组体重,水平活动和垂直活动得分,海马单胺类神经递质5-HT、NE和DA含量均显著增加,差异均有统计学意义(P〈0.05或P〈0.01);柴胡疏肝散中剂量组水平活动得分,5-HT和DA含量均显著增加,差异均有统计学意义(P〈0.05或P〈0.01)。结论中药柴胡疏肝散抗抑郁效果显著,可能与增加海马组织单胺类神经递质含量有关。

Abstract:Objective To explore the effect of Chaihu Shugan Powder (CSP) on content of hippocampus neurotransmitter in depression rats. Methods Sixty SD rats were randomly divided into were blank group, model group, fluoxetine group, medium-dose and high-dose CSP group, 12 cases in each group. Reserpine injection were intraperitoneal injected for continuousl4 days to establish model in all the groups except the blank group. After establishing, the fluoxetine group, medium-dose and high-dose CSP group were treated with 9mL/kg of fluoxetine, 5g/'kg and 10 g/kg of CSP respectively. After the 0, 4, 7, 10, 14 days of administration, the weight was recorded; after the 0, 7, 14 days, the score of Open-Field was calculated; the content of 5-hydroxytryptamine(5-HT), noradrenaline(NE) and dopamine (DA) of monoamine neurotransmitter in hippocampus tissue was detected by ELISA. Results Compared with the model group, the weight, horizontal and vertical score, and content of 5-HT, NE and DA in the high-dose CSP group were obviously increased, with significant difference( P 〈 0. 05 or P 〈 0. 01 ) ; horizontal score and the content of 5-HT and DA in the medium-dose CSP group were apparently increased, with significant difference (P 〈 0. 05 or P 〈 0. 01 ). Conclusion CSP has a good effect on anti-depression, which might be related with increasing the content of monoamine neurotransmitter in hippocampus tissue.
