亚砷酸钠对人胚胎肺成纤维细胞p53基因表达的诱导作用观察

摘    要:
目的探讨亚砷酸钠(NaAsO2)对体外培养的人胚胎肺成纤维细胞(HELF)p53基因表达的影响。方法以0、3、9、15μmol/L的NaAsO2处理体外培养的HELF24h,然后用Real-ti me PCR检测p53mRNA的表达,免疫组化SABC法检测p53蛋白水平。结果3、9、15μmol/L NaAsO2处理24h后,HELF中p53mRNA及蛋白表达均明显增高(P<0.05),且除9、15μmol/L组间p53mRNA表达无明显差异外,其余各组mRNA及蛋白表达水平均随NaAsO2剂量升高而增加(P<0.05)。直线相关分析显示,HELF中p53mRNA及蛋白表达均与NaAsO2剂量呈正相关(r=0.947,P<0.01;r=0.992,P<0.01)。结论NaAsO2可诱导HELF中p53mRNA和蛋白表达增加,并与剂量呈正相关。

Expression of p53 gene in human embryonic lung fibroblasts induced by sodium arsenite

Abstract:
Objective To study the dose-dependent effect of sodium arsenite on the expression of p53 gene in human embryonic lung fibroblasts (HELF) in vitro. Methods The human embryonic lung fibroblasts were divided into four groups in vitro based on completely randomized design. The expression of p53 mRNA was detected by real-time PCR and the expression of p53 protein was detected by immunohistochemical SAB, respectively, in human embryonic lung fibroblasts which were exposed to different doses (0, 3. 9 and 15μmol/ L) of sodium arsenite for 24 hours. The one-way analysis of variance and post hoc comparisons were performed for testing the differences among groups and the linear correlation was for testing correlation between doses and the expression of p53 gene. Results Compared with that of exposure to 0μmol/L sodium arsenite, the expression of p53 mRNA in HELF was increased significantly (P<0. 05) after being exposed to 3, 9 and 15μmol/L sodium arsenite for 24 hours. The significant difference (P<0. 05) was found between any two groups except that between 9 and 15μmol/L groups (P>0. 05). The p53 mRNA expression was increased in dose-dependent manner with the increased concentration of sodium arsenite (r=0. 947, P<0. 01). The expression of p53 protein in HELF was increased significantly (P< 0.05) after being exposed to 3, 9 and 15μmol/L sodium arsenite for 24 hours compared with that without sodium arsenite. Significant difference (P<0. 05) was found between any two groups. The p53 protein expression was increased in dose-dependent manner with the increased concentration of sodium arsenite (r=0.992, P<0.01). Conclusion Sodium arsenite can improve the expression of p53 gene in human embryonic lung fibroblasts in a dose-dependent manner.
