血管内皮生长因子在突出椎间盘重吸收中的表达及其意义

摘    要:目的研究血管内皮生长因子(VEGF)在正常椎间盘和不同突出类型、不同时间段内椎间盘突出后重吸收中的表达和意义。方法将63只雄性SD大鼠随机分为模型组Ⅰ(椎间盘不刺破组,模拟未破裂型突出)、模型组Ⅱ(椎间盘刺破组,模拟破裂型突出)、假手术组。并且取10 d、20 d、30 d三个时间段来观察。将所取得的大鼠椎间盘做VEGF免疫组织化学染色。结果对照组(假手术组)中椎间盘组织VEGF表达均为阴性,模型组VEGF的阳性表达均高于对照组(P〈0.05),其中,模型组ⅡVEGF的阳性表达高于模型组Ⅰ(P〈0.05);模型组ⅠVEGF的表达在各个时间段差异不大,模型组Ⅱ30 d和20 d VEGF的阳性表达均高于10 d的阳性表达(P〈0.05)。结论突出的椎间盘组织中有VEGF的表达,并且与突出类型和时间相关,后者可通过促进新生血管的形成,在腰椎间盘破裂型突出后重吸收中起促进作用。

Abstract:Objective To investigate the expression and significance of vascular endothelial growth factor(VEGF)in the spontaneous resorption in the various type of herniated lumbar intervertebral disc at different periods and the normal control.Methods Sixty-three male SD(Sprague dawley)rats were chosen.The rats were randomly divided into 3 groups:model group Ⅰ(Disc not puncture group,simulation unruptured prominent)、model group Ⅱ(Disc puncture group,simulated rupture prominent)、sham-operated group.And took 10 d、20 d、and 30 d different intervals observe the results.Intervertebral discs in rats made to do VEGF immunohistochemistry.Results There was no capillary infiltration and VEGF positive expression in the control group(sham-operated group),model group,the VEGF positive expression was higher than that in the control group(P0.05),them model group Ⅱ,VEGF positive expression was higher than that the model groupⅠ(P0.05);Model groupⅠVEGF expression is not different at all time,Model group Ⅱ30 d and 20 d VEGF positive expression was higher than 10 d in the positive(P0.05).Conclusion The herniated lumbar intervertebral disc tissue in the expression of VEGF,and associated with the prominent type and time,the latter can promote the formation of new blood vessels,in ruptured lumbar disc herniation in the post-accelerating effect on reabsorption.
