摘    要:目的研究氯胺酮对培养神经细胞兴奋性氨基酸N-甲基-D-天冬氨酸(NMDA)损伤的保护作用。方法采用原代培养的胎鼠皮层神经细胞模型,给予NMDA直接损伤,加入不同浓度的氯胺酮,测定神经细胞损伤后噻唑蓝(MTT)变化。结果氯胺酮在50~1000μmol/L范围内存在对兴奋性氨基酸损伤的保护作用,非剂量线性相关。结论氯胺酮对神经元NMDA兴奋毒性损伤产生拮抗作用,离体细胞模型中1×102-3μmol/L浓度左右具有保护效果。

Abstract:Objective To study the neuroprotective effect of ketamine for impairment in cultured neurocytes caused by NMDA.Methods By using primary cultured rat cortical neurons,the changes of MTT in neurons impaired by NMDA and protective effects of ketamine were studied.Results The neuroprotective effect of ketamine is closely related to the concentration of ketamine,the concentration between 50~1 000 μmol /L is its effective range and it is not related to linear dosage.Conclusion The neuroprotective effects of ketamine is operated in universality,and 1×102-3 μmol /L L is the best concentration for neuroprotective effect.
