原发性胆汁性肝硬化患者外周血IL15的表达及临床意义

目的检测IL15在原发性胆汁性肝硬化(primary biliary cirrhosis,PBC)患者中的表达情况,并初步探讨IL15与疾病发生发展关系。方法采用荧光定量PCR检测30例PBC、30例乙型肝炎肝硬化患者(疾病对照)和30例健康对照者的外周血单个核细胞(PBMC)中IL15 mRNA的表达,酶联免疫吸附试验(enzyme linked immunosorbent assay,ELISA)检测血清中IL15蛋白水平,并分析IL15与PBC患者肝功能指标之间的相关性。结果和健康对照组相比,PBC组和疾病对照组IL15 mRNA和蛋白的表达明显升高(P<0.05);PBC患者IL15的蛋白表达水平与总胆红素(TBIL)、r-谷氨酰基转移酶(r-GT)、碱性磷酸酶(ALP)水平均呈显著正相关(r=0.599,P<0.01;r=0.407,P<0.05;r=0.452,P<0.05)。结论IL15的表达与PBC的发生发展存在一定的相关性,可能与肝内胆管损伤及其严重程度有关,可作为PBC的辅助诊断和病情监测指标之一。

The Role of IL15 Expression in Patients with Primary Biliary Cirrhosis

Objective To evaluate the relation of IL15 expression with the development and progression patients with primary biliary cirrhosis(PBC).Methods Real time quantitive reverse transcription polymerase chain reaction(PCR)was used to detect the quantity of IL15 mRNA and Enzyme-linked immunosorbent assay(ELISA)was used to determine the protein levels of IL15 in patients with PBC(n=30),post-hepatitic cirrhosis(n=30) and in healthy control(n=30).Results Both the transcription and protein expression levels of IL15 were significantly higher in PBC patients and disease control,compared to healthy groups(P<0.05),and the expression of IL15 protein was positively correlated with TBIL,r-GT and ALP level respectively(r=0.599,P<0.01;r=0.407,P<0.05;r=0.452,P<0.05).Conclusion The expression levels of IL15 were correlated with the occurrence and development of PBC.Thus,IL15 may be of significance in the regulatory mechanisms governing PBC development,and therefore may be valuable in its diagnosis and prevention.
