银染法观察甲状腺激素人工神经促进周围神经再生的研究

摘    要:目的 用Bielschowsky神经纤维银染法观察再生有髓神经通过PDLLA T3 支架材料的情况及大鼠有髓再生神经纤维的数量。方法 SD大鼠 5 4只 ,分成PDLLA T3 、PDLLA组和自体移植组 3组 ,同只大鼠右侧坐骨神经为正常对照。培养新生大鼠坐骨神经的雪旺细胞 ,将其种植在PDLLA T3 和PDLLA材料上备用 ,每只大鼠左侧坐骨神经经手术造成 1cm缺损 ,然后分别用以上 3组材料进行移植修复 ,在伤后 2周、1个月、2个月 3个时相点收集标本进行神经银染 ,染色结果通过图象分析仪进行分析 ,统计数据行t检验。结果 伤后 2周时各组均见新生的神经纤维通过神经移植段 ,各组之间无显著性差异 ,直到伤后 1个月 ,2个月后 ,PDLLA T3 组好于PDLLA组 ,差异有显著性 ,P <0 .0 5。自体移植组髓神经增加明显 ,高于其他 2组 ,差异有显著性 ,P <0 .0 5 ,但低于正常值。结论 Bielschowsky神经纤维银染法观察大鼠神经再生效果满意 ,新生的神经纤维可以通过PDLLA T3 ,效果好于PDLLA。

Abstract:Objective To observe the number of regenerating myelinated nerve fibers that have passed through the scaffolds of PDLLA-T 3 by the method of Bielschowsky silver staining,which bridged the gap of 10-mm length in rats's left sciatic nerves.Methods Fifty-four SD rats were divided into 3 groups:PDLLA-T 3 group,PDLLA group and nerve autograft group.A sciatic nerve gap of 10-mm length in the left side was done by surgery in each rats,and the right nerve was collected as normal control.Schwann cells isolated from sciatic nerves of the new born rats were cultured for 2 weeks in vitro and then seeded onto the scaffolds of PDLLA-T 3 and PDLLA.The gaps in sciatic nerve were bridged with these scaffolds containing Schwann cells and nerve autograft respectively.Specimen were collected after 2 weeks,1 month,2months respectively and stained by the method of Bielschowsky silver staining,all the results were analyzed by image analyzer and then statistically treated by t-test.Results New myelinated nerve fibers were observed in the 3 groups after 2 weeks and 1 month,no significant difference was found among them.Two months later,more new fibers were observed in PDLLA-T 3 group than that in PDLLA group,the difference was significant,P<0.05,which was less than that of autograft group.Conclusion The scaffold of PDLLA-T 3 could bridge the nerve gap of 10mm successfully,the regenerating myelinated nerve fibers could pass through the PDLLA-T 3 scaffold,there were more new fibers than that in PDLLA.Bielschowsky silver staining is satisfactory method to observe new myelinated nerve fibers.
