胰岛素对人乳癌细胞株化疗敏感性的增强作用

目的:观察胰岛素能否增强人乳癌细胞株对化疗药物顺铂(DDP)的化疗敏感性.方法:体外培养的人乳癌细胞株(MDA-MB-543)分为8组,前7组分别于加入DDP前加入7.5 U/L的胰岛素作用3 h,6 h,9 h,12 h,15h,18 h,21 h,单纯DDP作用组(对照组)仅加入DDP.采用MTT法观察各组MDA-MB-543细胞生长的抑制率.结果:7.5 U/L的胰岛素作用9 h、12 h、15 h后加入DDP组MDA-MB-543细胞生长的抑制率明显高于DDP单独作用组,P均＜0.01.结论:胰岛素作用一定时间可以增强人乳癌细胞株对DDP的化疗敏感性.

Chemosensitization effect of insulin on cisplatin in vitro in MDA-MB-543 cells

Aim: To study the chemosensitization effect of insulin on chemotherapeutic drug cisplatin (DDP) in human breast cancer cells.Methods:The MDA-MB-543 cells were allocated into 8 groups, the 7 groups were given insulin at 7.5 U/L for 3 h,6 h,9 h,12 h, 15 h, 18 h, and 21 h,respectively before being given DDP, and the DDP group was only given DDP. The inhibition rate was calculated using MTT assay.Results: After being cultured with 7.5 U/L insulin for 9 h, 12 h, 15 h, respectively, the inhibition rate of DDP in DDP insulin groups was significantly higher than that of DDP group(P<0.01).Conclusion: Insulin can strengthen the therapeutic effects of DDP on human breast cancer cell line.
