缺血预适应处理对大鼠移植肺组织中HSP70 mRNA表达的影响

摘    要:
目的:应用两套管-支架管大鼠肺移植模型,观察缺血预适应对大鼠移植肺组织中HSP70 mRNA表达的影响.方法:60只大鼠随机分为3组(每组按肺移植受、供体配成10对):对照组应用两套管-支架管法建立大鼠左肺移植模型;缺血预适应组供体大鼠开胸后先阻断左肺动脉和左主支气管5 min,然后开放10 min,造成左肺(移植肺)的缺血预适应;预适应阻断组供体大鼠在缺血预适应后,经肺动脉灌注蛋白激酶C的阻断剂多黏菌素B.分别于移植成功后30 min,自左肺静脉抽取1 mL动脉血即送血气分析,记录氧分压;采用黄嘌呤氧化酶法测定SOD活力;应用RT-PCR法检测移植肺组织中HSP70 mRNA的表达强度.结果:缺血预适应组的左肺静脉血氧分压、SOD、HSP70 mRNA均高于预适应阻断组和对照组,差异有统计学意义(P<0.05).结论:缺血预适应能够增强大鼠移植肺组织中HSP70mRNA的表达,提高SOD的活力,对移植肺有保护作用.

Abstract:
Aim: To investigate the effect of ischemic preconditioning on the expression of HSP70mRNA in rat lung tissue using two-cuff-one-lung transplantation model. Methods: Sixty SD rats were randomly divided into three groups and establised transplantation model, respectively: control group (the donor lung was preserved normally), preconditioning group (the donor lung underwent ischemic preconditioning before preservation) and blockade group (specific blocking agent of protein kinase C, polymyxin B, was infused via pulmonary artery after ischemic preconditioning), respectively. pO2 of left pulmonary vein, HSP70 mRNA and SOD activity were detected when the donor lung were anastomosed. Results: pO2 , HSP70 mRNA and SOD activity in preconditioning group were significantly higher than those in control and blockade groups (P<0.05). Conclusion: Ischemic preconditioning can strengthen the expression of HSP70 mRNA, and improve the activity of SOD, thus have protective effects on transplanted lung.
