马红丸对肝癌细胞粘附迁移侵袭能力的影响

摘    要:
目的 利用马红丸含药血清研究其对SK-hep-1细胞的粘附迁移侵袭能力的影响。方法 SK-hep-1细胞分别用10%(V/V)、20%(V/V)、30%(V/V)马红丸含药血清、阿霉素含药血清、正常对照组(10%胎牛血清)培养,并观察SK-hep-1细胞的迁移粘附侵袭能力,采用SPSS16.0软件进行数据处理及统计学分析,各组之间采用两独立样本资料的t检验。结果 10%(V/V)、20%(V/V)马红丸含药血清组细胞粘附个数均值分别为(39.00±6.51)和(38.00±2.00),与正常对照组(90.30±6.11)相比,差异有统计学意义(P0.01);10%(V/V)、20%(V/V)马红丸含药血清组迁移个数均值分别为(6.00±3.61)和(12.33±3.79),与正常对照组(57.67±5.69)相比,差异有统计学意义(P0.01);10%(V/V)、20%(V/V)马红丸含药血清组细胞侵袭个数均值分别为(70.03±9.39)、(41.77±12.04),与正常对照组(100.00±9.74)相比差异有统计学意义(P0.05或P0.01);30%(V/V)马红丸含药血清作用SK-hep-1细胞,与正常对照组相比差异无统计学意义(P0.05),20%马红丸含药血清抑制效果最好。结论 马红丸能够有效抑制肝癌细胞的细胞运动能力,从而减缓肝癌转移,本研究利用现代分子生物学技术阐明该药减缓肝癌侵袭转移的分子机制,为马红丸进一步临床应用提供良好的理论基础。

Abstract:
Objective To study the effect of Mahong pill (a traditional Chinese patent medicine) on adhesion,migration and invasion of SK-hep-1.Methods SK-hep-1 cells were cultured in 10% (V/V),20% (V/V),30% (V/V) Mahong pill containing medicine serum,adriamycin containing medicine serum,and normal control group (10% fetal bovine serum),and observed SK-hep-1 cells migration,adhesive ability.SPSS16.0 software was used for data processing and statistical analysis,by using two independent samples t test.Results The average cells of adhesion in 10% (V/V) and 20% (V/V) Mahong pill containing medicine serum group were (39.00 ± 6.51) and (38.00± 2.00),respectively,compared with normal control group (90.30±6.11),the difference was statistically significant (P<0.01);The mean number of migration cells in 10% (V/V) and 20% (V/V) Mahong pill containing medicine serum group were (6.00±3.61) and (12.33±3.79),respectively,compared with normal control group (57.67±5.69),the difference was statistically significant (P<0.01);The mean number of invade cells in 10% (V/V) and 20% (V/V) Mahong pill serum group were (70.03±9.39),(41.77± 12.04),respectively,compared with normal control group (100.00±9.74),the difference was statistically significant (P<0.05 or P<0.01);SK-hep-1 cells in 30% (V/V) Mahong pill containing medicine serum was no statistically significant difference,compared with normal control group (P>0.05),inhibition effect of 20% Mahong pill containing medicine serum was best.Conclusion Mahong pill can effectively restrain the cancer cell movement ability,ability to slow liver cancer metastasis,this study using modern molecular biology techniques to clarify the drug slow the molecular mechanisms of invasion and metastasis of liver cancer,for further provide a good theoretical basis for clinical application of Mahong pill.
