针刺对肥胖大鼠下丘脑神经肽Y及脂肪蓄积的影响

目的:观察针刺对肥胖大鼠下丘脑神经肽Y及脂肪蓄积的影响,并探讨针刺减肥的治疗机理。方法:将60只SD大鼠造模后随机分为正常对照组、肥胖对照组及针刺治疗组,观察各组大鼠针刺治疗前、后的体重及Lee's指数。针刺治疗后取各组大鼠下丘脑常规免疫组织化学法检测神经肽Y表达,取脂肪组织染色后体视学方法测量体密度、数密度及表面积密度。结果:治疗后,针刺治疗组体重、Lee's指数与正常对照组无差异,与肥胖对照组相比明显减低(P<0.01);针刺治疗组下丘脑NPY积分光密度及脂滴体密度、表面积密度、数密度与肥胖对照组相比明显下降(P<0.01),与正常对照组相比无差异。结论:针刺治疗肥胖效果显著,且显著降低肥胖大鼠升高的下丘脑NPY含量,减少肥胖大鼠已增加的脂肪蓄积量。

Influence of acupuncture on hypothalamic Neuropeptide Y and lipopexia of obese rat

Objective To observe the influence of acupuncture on hypothalamic neuropeptide Y and lipopexia of obese rat, and to explore the therapeutic mechanism of weight reduction by acupuncture. Methods Sixty SD rats were randomly divided to three groups: normal control, obese control and acupuncture treatment. Then the weight and Lee's Index of all rats were measured. After acupuncture treatment, the hypothalamus of all rast were fixed for immunohistochemical staining of neuropeptide Y, and the bulk density, number density and surface area density of lipid droplet were measured. Results Compared with obese control, the weight and Lee's Index of rats in acupuncture treatment were decreased markly, and the integral optical density of hypothalamic neuropeptide Y and the bulk density, number density and surface area density of lipid droplet were down-regulated obviously. Conclusion Weight reduction by acupuncture is effective. In addition, the up-regulated expression of hypothalamic neuropeptide Y and the up-regulated lipopexia could be decreased significantly by acupuncture
