大鼠鼠胚海马神经元缺氧复氧致细胞凋亡模型的建立

摘    要:目的建立大鼠鼠胚海马神经元缺氧复氧致细胞凋亡模型。方法在大鼠鼠胚海马神经元培养基础上,给予不同时间的缺氧复氧,应用Wright-Giemsa染色、TUNEL法检测细胞凋亡情况。结果随着缺氧和复氧时间的延长神经细胞凋亡数逐渐增加,其中缺氧4 h复氧48 h大鼠海马神经细胞凋亡最为显著。结论缺氧复氧可诱导培养的大鼠海马神经细胞发生凋亡,缺氧4 h复氧48 h可作为细胞凋亡模型。

Abstract:Objective:To establish hippocampal neuron apoptosis model induced by hypoxia-reoxygenation.Methods:On the basis of culturing hippocampal neurons of embryo of rat,neurons were given different hypoxia-reoxygenation time and apoptosis was studied by using methods of Wright-Gimsa staining,TUNEL.Results:The percentage of apoptotic neurons was found to be increased along with the prolongation of hypoxia-reoxygenation.Apoptosis of neurons undergoing 48 hours reoxygenation after 4 hours of hypoxia was mostly significant.Conclusion:Hippocampal neuron apoptosis may be induced by hypoxia-reoxygenation and cultured hippocampal neurons undergoing 48 hours reoxygenation after 4 hours of hypoxia can be used as an available cell model of hippocampal neuronal apoptosis for futher studies.
