高聚金葡素对非小细胞肺癌细胞株A549的生长抑制及其治疗恶性胸腔积液的效果观察

摘    要:目的:观察生物调节剂高聚金葡素(high agglomerative staphylococoin,HAS)对非小细胞肺癌细胞株A549的体外生长抑制,并观察HAS治疗肺癌恶性胸腔积液的临床疗效。方法:培养非小细胞肺癌细胞株A549,根据处理方法不同,分为对照组、顺铂(cisplatin,DDP)组、HAS、HAS联合DPP 4组,应用CCK8法测定药物的半数抑制浓度(50%concentration of inhibition,IC50)值及细胞存活率,倒置显微镜下观察各组细胞形态改变。收集非小细胞肺癌胸腔积液患者的胸膜腔积液,并应用HAS、DDP或两者联合进行治疗,观察其临床疗效和不良反应。结果:HAS能抑制肺癌细胞株的生长,并且呈时间剂量依赖性,HAS联合DDP治疗较DDP单独治疗IC50值明显降低。在对临床恶性胸腔积液患者的治疗观察中,HAS治疗有效率为72.2%,DDP组治疗有效率为50.0%,HAS联合DDP治疗有效率为86.1%,且HAS组发生胃肠道反应和骨髓抑制的比例要小于DDP组。结论:HAS对非小细胞肺癌具有生长抑制作用,局部应用HAS不良反应小,能有效促进恶性胸腔积液的吸收。

Abstract:Objective: To investigate the effects of high agglomerative staphylococoin(HAS) on the growth of non-small cell lung cancer cell line A549,and to evaluate the effect of HAS in the treatment of malignant pleural effusion.Methods: A549 cells were incubated in F12-K medium containing 10% fetal bovine serum,2.5 μg/mL fungizone and 100 U/mL streptomycin/penicillin,then they were treated with different concentration of HAS and DDP.CCK-8 assay was used to measure the inhibitory rate.Phase contrast microscope was used to observe the morphological changes of A549 cells treated with HAS.One hundred and eight lung cancer patients with malignant pleural effusion were randomly divided into three groups,and treated with DDP,HAS,DDP and HAS,respectively.Results: Both HAS and DDP could inhibit A549 cell proliferation in time-dependent and dose-dependent manners.Treating with HAS and DDP together decreased the IC50 of DDP than treating with DDP alone.The overall response of HAS group was 72.2%,which was higher than that of DDP group(P0.05).Conclusions: HAS,which is less toxic than DDP,can inhibit proliferation of A549 cells.Therapy of HAS can effectively control the malignant pleural effusion and improve life quality of the lung cancer patients.
