新洁尔灭对小鼠骨髓嗜多染红细胞微核的影响

摘    要:
新洁尔灭从70年代末以来被用于终止早期妊娠,据资料记载其有直接杀胚和溶血作用。本实验用NIH小鼠骨髓嗜多染红细胞微核测定的方法,对新洁尔灭进行了是否有致突变作用的检测,采用腹腔注射LD_(50)的1/8~1/2量,一次性腹腔注射,另设阴性(生理盐水)的阳性(环磷酰胺)对照。结果,阴性对照组的骨髓PCE微核细胞率为1.25±0.83,微核率是1.25±0.83,新洁尔灭低剂量组为4.0±1.5和4.7±2.5,中剂量组为2.8±0.7和3.0±0.6,高剂量组为5.2±1.7和6.2±1.9。阳性对照为12.6±5.0和14.6±6.48。经统计学处理,低、中、高三个剂量组与阴性对照组之间均有显著性差异,因而从本实验结果来看,新洁尔灭可能具有诱变作用,提请广大医务工作者在用于抗早孕时务必注意其安全性。

Effect of Bromo-Geramine(G-12)on Micronuclei of Polychromatophilic Erythroblasts in Mice.
Abstract:
Fifteenth years ago. Bromo-Geranime (G-12) was used for the termination of eraly pregnancy. It was reported that the a abortifacient effect is due the damage of embryo directly and with hemolytic effect. The present investigation evaluated the effect of Bromo-Geramine (G-12) by means of observing the micronuclei of polychromatophilic erythorbast exchange in bone marrow of NIH mice,by adminis-tration of normal salione and cyclophophamide as negative and positive control group. The results showed that the frequency of occurrence of mi-cronucleated polychromatophilic erythrobasts and frequency of micronucle as follows respectively: 1. 25±0. 83 and 1. 25±0. 83 (negative group), 4. 0 ±1.5and 4. 7 ±2. 5(G-12small dosage group), 2. 8 ±0. 7and3. 0±0. 6(G-12middle dosage group),5. 2 ± 1. 7 and 6. 2 ± 1. 9 (G - 12 maximum dosage group), 12. 6±5.0andl4. 6±6. 48(positive group). Three G-12groups compared separatel with those of the control group,there were significant differences between them(P<0. 05 P<0. 001) It is suggested that the drug might induce the mutation of somatic cells and it should the used carefully.
