冠心合剂减少大鼠心肌梗死边缘区凋亡及对Fas、FasL、Caspase-8、Caspase-3蛋白表达的影响

摘    要:目的:观察不同剂量冠心合剂对大鼠急性心肌梗死边缘区细胞凋亡及探讨可能作用途径。方法:取雄性SD大鼠80只,随机分为模型组、冠心合剂高剂量组、中剂量组、低剂量组和对照组。连续灌胃7d后,结扎冠状动脉左前降支造成急性心肌梗死模型,对照组不结扎冠状动脉左前降支,手术后4.5h留取心脏标本。检测梗死边缘区细胞凋亡以及Fas,FasL,Caspase-8,Caspase-3蛋白表达量。结果:与模型组比较,冠心合剂组可明显减少心肌细胞凋亡,并能减少Fas,FasL,Caspase-8,Caspase-3的蛋白表达量。其中冠心合剂大剂量组减少心肌细胞凋亡效果最明显。结论:冠心合剂能够保护缺血诱导的心肌细胞凋亡,其作用机制Fas,FasL,Caspase-8,Caspase-3蛋白表达水平有关。

Guanxin Heji Reducing Apoptosis in Rats with Acute Myocardial Infarction and Effects on Fas, FasL, Caspase - 8, Caspase - 3 Expressions

Abstract:Objective:To investigate the effects of different dosages of Guanxin Heji on apoptosis in rats with acute my- ocardial infarction and its possible mechanism. Methods : 80 male SD rats were randomly divided into 5 groups : model group, sham group, high - dose group [ 28 g/( kg. d ) Guanxin Heji pretreatment ], medium - dose group [14 g/( kg . d ) Guanxin Heji pretreatment ], low - dose group [ 7 g/( kg .d ) Guanxin Heji pretreatment ]. After being administered corresponding medicine for 7 days, all groups except sham group underwent coronary ligation of left anterior descending branch to establish acute myocardial infarction models, and sham group only received thoracotomy without cordnary liga- tion. Four and half hours after acute myocardial infarction, myocardium tissues were collected to detect the apoptotic index with TUNEL and protein expressions of Fas, FasL, Caspase - 8 and Caspase - 3 by Western blot. Results : The apoptotic index and protein expression sof Fas, FasL, Caspase -8 and Caspase -3 were decreased significantly in all dose Guanxin Heji groups than those in model group. The high - dose group received the best effect than the others. Conclusion : Guanx- in Heji inhibited the myocardial apoptosis by reducing apoptosis through Fas/FasL pathway.
