Gas2基因在肝细胞癌中的表达及意义

目的　探讨Gas2基因在HCC中的表达规律及与HCC中细胞凋亡和增殖的关系。方法　对 36例癌、癌旁肝组织及 8例正常肝组织 ,应用石蜡切片原位杂交技术观察Gas2mRNA在HCC中的表达。TUNEL技术检测凋亡细胞 (APOP) ,免疫组织化学方法检测PCNA的表达作为细胞增殖的参数。结果　肝癌、癌旁及正常肝组织中均有半数以上病例有Gas2mRNA表达 ,但肝癌组阳性积分显著高于癌旁和正常肝。Gas2mRNA表达高的组织内见较多凋亡细胞 ,以不同病理特征分组中 ,Gas2mRNA阳性积分与APOP阳性分值多呈现一致性。Gas2mRNA表达与细胞增殖无明显相关。结论　Gas2mRNA在癌内表达明显较高 ,其阳性表达与细胞凋亡关系较密切 ,Gas2基因有可能通过影响细胞的凋亡状况而在HCC的生长过程中起作用。

Gas2 Gene Expression and Its Significance in Hepatocellular Carcinoma

Purpose To investigate the possible effect of the Gas2 gene in HCC. Methods Using the techniques of in situ hybridization for RNA?RNA in paraffin sections to detect Gas2 mRNA,TUNEL to find out the apoptotic cell and immunohistochemistry of PCNA to present the proliferating cells in both cancerous lesion and non-cancerous tissue of the resecting specimens of 36 cases of HCC as well as 8 cases of normal liver tissues. Results Gas2 mRNA signals present in more than 50% cases of cancerous,non-cancerous and normal liver tissues,with the highest expression in the cancerous tissues.More apoptotic cells can be seen in the tissues with stronger Gas2 mRNA expression.In most of cases classified by varied pathologic features,Gas2 value integral appears to be generally identical to A .There is no significant relationship between the expression of Gas2 mRNA and cell proliferation(PCNA expression). Conclusions Expression of Gas2 mRNA in the cancerous tissue is much higher.There seems close relationship between the expression of Gas2 gene and cell apoptosis by which Gas2 may play some role on the growth of HCC.
