摘    要:
本实验观察了脑室注射血管紧张素I(AI)对成年雄性Wistar大鼠下丘脑促性腺激素释放激素(GnRH)含量的影响。摘取下丘脑组织,匀浆化,用放射免疫分析检测匀浆上清中GnRH的含量。结果表明,脑室注射AI(120ng/20μl)后60、90及120分钟,下丘脑GnRH含量分别为2.88±0.47、2.69±0.38及2.54±0.55ng/10mg湿重组织,均显著高于盐水对照组(P<0.05),提示AI可促进雄性大鼠下丘脑GNRH的生成。

Abstract:
Objectives: To investigate the effect of intracerebroventricular injection of angiotensin II on hypothalamic gonadotrophin releasing hormone (GnRH) production in male rats. Design: Experimental animal study. Settings: Harbin Medical University. Methods: Male rats were anaesthetized with ether, then the saline (20 μl) and A II solution (at 30, 60 and 120 ng/20 μl) were injected intracerebroventricularly. The hypothalamic tissues were removed at 60, 90 and 120 min after intracerebroventricular injection of A II, followed by homogenization and centrifugation. The GnRH content in supernates of hypothalamic homogenates were measured by RIA. Results: The mean hypothalamic GnRH contents were 2.88±0.47, 2.69±0.38 and 2.54±0.55 ng/10mg wet tissue at 60, 90 and 120 min after intracerebroventricular injection of 120 ng A II, which were significantly higher than those in saline control groups (P<0.05). Conclusions: These results demonstrated that A II can stimulate GnRH production in the hypothalamus of male rat.
