金菊双花总黄酮提取与纯化工艺研究

摘    要:目的:筛选金菊双花总黄酮的最佳提取与纯化工艺.方法:采用多种提取工艺比较、L9(34)正交实验及D101大孔吸附树脂纯化技术.结果:其最佳提取工艺为8倍量55%乙醇、100℃水浴、每次1 h回流提取2次,再结合D101大孔吸附树脂纯化,水、30%乙醇、70%乙醇梯度洗脱,收集70%乙醇洗脱部分浓缩干燥并测定黄酮含量为62.7%.结论:此工艺简便可行,符合工业化生产要求.

Abstract:OBJECTIVE: To optimize the extraction and purification conditions for total flavonoids in Lonicera japonica Thunb. and Chrysanthemum morifolium Tzvel. METHODS: Comparision of several extraction processes, the L9 (3(4)) orthogonal design and macroreticular absorption resin D101 purification technique were performed. RESULTS: The optimum extraction conditions were 8 volume of 55% ethanol, 1 h refluence at 100 degrees C , water bath for 2 times. The extracted fluid was absorbed with the resin D101. The elutions were water, 30% enthanol, 70% enthanol. The obtained elution of 70% enthanol was evaporated and the content of total flavonoids was 62. 7%. CONCLUSION: The method is simple, approved and fit for industrial production.
