哮喘大鼠肺组织白血病抑制因子的表达及地塞米松干预研究

目的探讨哮喘大鼠肺组织中白血病抑制因子(leukemia inhibitory factor,LIF)的表达及其意义.方法制作卵蛋白致敏大鼠哮喘模型,采用RT-PCR和Western-blot方法观察LIF在致敏大鼠肺组织中的表达.结果LIF蛋白表达量在哮喘组为(40832±12964),均较地塞米松组(16160±2108)和对照组(23110±8018)升高,差异有显著性(P＜0.01),地塞米松组与对照组之间无显著性差异(P＞0.05).LIFmRNA的相对表达量哮喘组为(0.51±0.13),均较地塞米松组(0.18±0.05)和对照组(0.24±0.02)明显增高,差异有显.著性(P＜0.01),地塞米松组与对照组之间无显著性差异(P＞0.05).结论LIF在致敏哮喘大鼠模型肺组织中表达显著增高,LIF作为神经免疫调节因子,可能参与了哮喘的发病机制.

Expression of leukemia inhibitory factor in lung in ovalbumin sensitized and dexamethasone therapied rats

Objective:To investigate the expression of leukemia inhibiObjective:Tory facObjective:Tor(LIF)in lung of ovalbumin sensitized and challenged rats for evaluating the role of LIF in pathogenesis of asthma.Methods:Asthma rat model sensitized and challenged by ovalbumin was established.The expression of LIF at mRNA and protein levels in the lung was detected by RT-PCR and western blotting.Results: Expression of LIF protein in the lungs of asthma rats(40832 ± 12964)was significantly elevated compared with control(23110 ± 8018,P＜0.01)and dexamethasone therapy(DXM)rats(16160 ± 2108,P＜0.01),but no difference was found between control group and DXM group(P ＞ 0.05).Expression of LIF mRNA in lungs of asthma rats(0.51 ± 0.13)was significantly elevated compared with control(0.24 ±0.02,P＜0.01)and DXM rats(0.18 ±0.05,P＜0.01),but no difference was found between control and DXM rats also(P ＞ 0.05).Conclusions: The expression of LIF at protein and mRNA levels are elevated significantly in ovalbumin sensitized rats,which indicates that LIF may play an important role in pathogenesis of asthma as a neuroimmune modulator.
