RP-HPLC法测定大鼠血清中龙胆苦苷的含量

目的　建立一种测定大鼠血清中龙胆苦苷含量的方法。方法　针对化合物的理化性质 ,采用固相萃取小柱对样品进行测定前的预处理 ,然后以甲醇 -水 (30∶ 70 )为流动相于高效液相色谱仪上定量 ,检测波长 2 70nm。结果　方法学考察结果显示 ,线性范围为 0 .5～ 15 6 .2 mg· L-1,且相关系数良好 (r =0 .9994 ) ,高、中、低浓度样品的回收率分别为 83.5 % ,81.9% ,84 .9% ,检测限小于 0 .13mg· L-1。结论　此方法操作简便 ,可作为一般药理学研究测定血清中龙胆苦苷浓度的方法 ,也可为临床监测人血清中龙胆苦苷浓度提供参考

Determination of gentiopicroside in rat serum by RP-HPLC

Objective To develop an HPLC method for the determination of gentiopicroside in rat serum. Methods A solid phase extraction(SPE) column was selected for pre dealing with the samples. The qualitative determination of them was done with a YWG ODS C 18 column, a detector set at 270 nm and methanol water(30∶70) at a flow rate of 1 mL·min -1 as mobile phase. Results A linearity was obtained from 0 5 to 156 2 mg·L -1 of gentiopicroside in serum with a good correltion coefficient(r=0 999 4, n=10). The mean recoveries were 84 9%, 81 9% and 83 5% for the low, middle, high concentrations of the samples respectively. The limit of detection was less than 0 13 mg·L -1 . Conclusion The method was sensitive, specific and simple. It is suitable for the pharmacokinetic study in rats.
