环氧合酶-2抑制剂对人胃癌细胞增殖及侵袭力影响的研究

目的 研究环氧合酶-2抑制剂SC236对人胃癌细胞株增殖及侵袭能力的影响，探讨环氧合酶-2抑制剂用于胃癌转移治疗的可能性。方法 人胃癌细胞株SGC7901经环氧合酶-2抑制剂SC236处理后，用MTT法检测其增殖能力的变化。用黏附基质分析法和Transwell法检测其侵袭能力的变化。结果 经SC236处理后，SGC7901的增殖受到抑制，细胞侵袭能力下降，黏附率下降。运动能力减低。结论 SC236能有效抑制胃癌细胞的生长。明显减低其侵袭能力。环氧合酶-2抑制剂有望成为治疗胃癌转移的有效药物。

Study on effect of cox-2 inhibitor on proliferation and invasion of human gastric cancer cells

Objective To explore the effect of cox-2 inhibitor on the proliferation and invasion of human gastric cancer.Methods After treatment of SGC7901cells with SC236,MTT assay was used to test the effect on the proliferation.Cell biologic behaviors such as adhesiveness,cell motility and in vitro invasiveness were observed.Results The suppressive effect of SC236 on the proliferation of SGC7901 is dependent on time and concentration.The ability of SC236 treated SGC7901 cells to invade the reconstituted basement membrane decreased(P<0.01),the cell motivity and attachment to fibronectin of drug-treated cells decreased too.Conclusion SC236 can inhibit the proliferation and invasion of human gastric cancer cells,cox-2 inhibitor might be a candidate drug for the prevention of metastasis of human gastric cancer.
