摘    要:
目前面神经损伤修复的机制尚未阐明,揭示面神经损伤后神经修复再生的病理生理机制,最大程度地恢复面神经功能,是学者们面临的一个重要问题。在前期研究中,本课题组发现毒蕈碱受体和烟碱受体通过不同的机制调控面神经核细胞内游离钙离子的水平。并且面神经损伤后,毒蕈碱受体和烟碱受体通过不同机制使面神经核游离钙离子浓度达到更高水平,为面神经的再生修复的一系列反应提供了重要条件。但是,细胞内钙离子水平增高到一定程度可以使细胞发生钙超载而死亡。γ-氨基丁酸(GABA)是中枢神经系统重要的抑制性神经递质,GABAA受体在面神经再生修复中对胆碱能受体效应的调控及机制目前仍为一空白领域。为了探讨GABAA受体在面神经损伤修复中的作用,本研究中建立面神经损伤动物模型,通过免疫组织化学、原位杂交和激光共聚焦显微镜技术首次系统研究了大鼠面神经核中GABAA受体的表达以及面神经损伤后GABAA受体对烟碱受体效应的调控。研究发现在面神经核至少存在GABAA的α1、α3、α5、β1、β2、δ、γ3亚单位。并且面神经损伤后GABAA受体对烟碱受体介导的面神经核游离钙离子水平增高的效应有显著的抑制效应(P<0.05),呈浓度依赖性。因此,通过本课题组一系列研究,我们认为周围性面神经损伤后,面神经核游离钙离子浓度至少受到毒蕈碱受体、烟碱受体的正向调控和GABAA受体的负向调控,从而为面神经再生修复提供合适的钙离子水平。
