摘 要: | 目的研究原发性肝癌(PHC)组织高迁移率族蛋白B1(HMGB1)表达改变和诱导机制,及PHC患者血清HMGB1水平变化和意义。方法对38例慢性乙肝、32例乙肝后肝硬化、23例继发性肝癌、39例PHC患者和48例健康对照者的血清HMGB1水平进行测定和分析。采用RT-PCR方法,检测11例PHC组织及其癌旁组织中的HMGB1基因表达水平。观察缺氧培养对肝癌细胞株SMMC-7721HMGB1基因表达和胞外释放的影响。结果PHC患者血清HMGB1水平(13.5±6.3μg/L)明显高于健康对照者(3.9±1.4μg/L),并与AFP水平、TNM分期有关。PHC组织中HMGB1mRNA表达增强。SMMC-7721细胞经缺氧培养3h后,培养上清液中HMGB1含量即见升高,6h后,HMGB1mRNA表达明显增强(P〈O.01)。结论PHCHMGB1表达增强,缺氧为诱导因素。

Abstract: | Objective To investigate the change of high mobility group box 1 (HMGB1) expression in primary he- patic carcinoma(PHC) tissues, its induction mechanism, serum HMGB1 levels in PHC patients and its significance. Methods serum HMGB1 levels were measured and analysed in 48 healthy control,38 chronic hepatitis,32 liver cirrho- sis,23 secondary hepatocarcinoma and 39 PHC. HMGB1 gene expression was analysed in 11 PHC tissues and their noncancerous tissues using RT-PCR. Using hepatocarcinoma cell SMMC-7721, the effect of hypoxia.on HMGB1 mRNA expression and HMGB1 release was observed. Results Serum HMGB1 levels in PHC patients (13.5±6.3 μg/L) were significantly higher than that in healthy controls (3.9 ±1.4 μg/L), and related with AFP concentration and TNM stage. HMGB1 mRNA expression in PHC tissues markedly raised. HMGB1 concentration in the culture supernatant in- creased at 3 h,and HMGBt mRNA expression enhanced at 6 h (P〈0. 01) ,after SMMC-7721 cells were subjected to hypoxia culture. Conclusion HMGB1 expression in PHC significantly increases,and hypoxia may be involved with its mechanism. |
