抗人DR5单抗对人肝癌细胞系的凋亡作用

摘    要:
目的:探讨抗人DR5单克隆抗体(mDRA-6)对人肝癌细胞系的凋亡诱导活性。方法:常规培养人肝细胞HL7702,单克隆抗体对细胞的杀伤活性用MTT法检测、对细胞的凋亡作用采用Hoechst33258染色荧光显微镜下观察细胞形态变化及Annexin V-FITC/PI双染流式细胞术定量分析技术。结果:MTT的结果显示抗体的量和细胞杀伤率之间有明显的线性关系,随着抗体浓度的增加,细胞的杀伤率就增加;荧光显微镜下mDRA-6诱导导致细胞呈现典型的细胞凋亡的形态性特特征;流式细胞术检测显示,2 mg/L的mDRA-6作用人肝肝癌癌细胞系SMMC-7721细胞6 h,细胞凋亡率为35%。结论:mDRA-6能够诱导人肝癌细胞系凋亡,mDRA-6具有诱导细胞凋亡的活性。

Abstract:
Objective:To evaluate the effect of a novel anti-human DR5 monoclonal antibody(mAb mDRA-6) on the apoptosis of human hepatocellular carcinoma cell lines.Methods: mDRA-6 cytotoxicity on cell lines was detected by MTT analysis.The influence of mAb mDRA-6on the morpha of cells lines was observed under fluorencence microscope.The rate of apoptosis was detected by flow cytometry with Annexin V-FITC/PI staining.Results: Cells treated with mDRA-6 exhibited typical apoptosis features in morphology.Viability of cells decreased with mAb concentration increasing by MTT analysis.The apoptosis rate of the SMMC-7721 cells was 35% in the presence 2 mg/L mDRA-6 for 6 hours by flow cytometry analysis with Annexin V-FITC/PI staining.Conclusion: mDRA-6 could induce SMMC-7721 cells apoptosis and has apoptosis activity.
